Raman spectroscopy measures the inelastic scattering of photons from a substance to retrieve information on vibrational modes, comparable to information obtained from infrared spectroscopy. The resonance frequency of vibrational modes depends on the masses of atoms, the bond strength and on interactions with the molecular environment. This makes Raman spectroscopy a powerful tool to identify chemicals based on their functional groups, study molecular bonding and probe intermolecular interactions, e.g., hydrogen bonding.
The Witec Alpha 300RAS is a confocal Raman microscope that combines the spatial resolution of optical microscopy with the chemical information gathered by Raman spectroscopy. This enables Raman spectroscopy measurements at selected points or areas across the sample surface with a lateral resolution down to the µm range. The Raman spectra can be overlaid with the respective optical microscopy image to achieve both chemical and optical contrast, which enables deep insights into complex biological system and material properties/engineering.
The Raman microscope offers the possibility to enhance the spatial resolution using the built-in atomic force microscopy (AFM) or scanning near-field optical microscopy (SNOM) down to the nanometer range.
